Project Description: The proposed project aims to investigate the potential impact of membrane fouling on downstream unit operations in IOPS. Many downstream filtration operations are susceptible to membrane fouling, including harvest, virus filtration, ultrafiltration and diafiltration (UF/DF) and sterile filtration. Membrane fouling caused by protein aggregation can cause process inconsistency and increase production cost. There is also a lack of understanding on the fouling mechanism and the associated impacts especially when the aggregate level is beyond normal manufacturing range. A generic aggregate spiking strategy is also needed to evaluate the impact for different downstream processes. This project will focus on the downstream unit operations that are sensitive to aggregates and investigate the impacts as well as the fouling mechanism.

Business benefit/advantage for IOPS: The information that will be gathered from this proposed project will address gaps in the understanding of filtration-based downstream operations. This proposed study will look deep into the fouling mechanism and evaluate the impact on process and product. The information from this study will be useful for providing the further understanding of membrane fouling on downstream operations in IOPS and can be helpful during studies into process investigations.
